Drug Target Encyclopedia Series (38) —DLL3

DLL3, also known as **Delta-like Ligand 3**, is an inhibitory Notch pathway ligand with limited expression in most normal adult tissues but high expression in several neuroendocrine tumors. Due to its tumor-selective expression profile, DLL3 has become an important target in oncology drug development, especially in small cell lung cancer.

DLL3 is highly relevant in small cell lung cancer (SCLC), large cell neuroendocrine carcinoma, neuroendocrine prostate cancer, and other neuroendocrine tumor research. Its cell-surface expression makes it particularly attractive for antibody-based and targeted therapeutic strategies.

🧬 DLL3 is highly relevant in:

• Small cell lung cancer

• Large cell neuroendocrine carcinoma

• Neuroendocrine prostate cancer

• Gastroenteropancreatic neuroendocrine tumors

• High-grade neuroendocrine carcinomas

• Solid tumor research

🎯 Targeting DLL3 in drug development:

🧪 Bispecific antibodies / T-cell engagers – Redirect T cells to DLL3-positive tumor cells and drive immune-mediated tumor killing.

🧪 Antibody-drug conjugates – Deliver cytotoxic payloads selectively to DLL3-expressing tumor cells.

🧪 CAR-T cell therapy – Explore DLL3 as a tumor-associated antigen for solid tumor cell therapy.

🧪 Radioligand and imaging strategies – Support patient selection, tumor imaging, and targeted radiotherapeutic development.

💥 Challenges include:

• Tumor antigen heterogeneity

• Managing on-target/off-tumor safety

• Solid tumor microenvironment barriers

• Optimizing patient selection

• Achieving durable responses in aggressive tumors

🔬 DLL3 continues to gain strong attention as a promising membrane protein target in neuroendocrine oncology. With progress in bispecific antibodies, ADCs, CAR-T therapies, and precision targeting strategies, DLL3 may play an important role in expanding treatment options for SCLC and other neuroendocrine tumors.
