STEAP1, also known as Six-Transmembrane Epithelial Antigen of the Prostate 1, is a tumor-associated cell surface antigen highly expressed in prostate cancer and several other solid tumors. With its multi-pass membrane structure and relatively restricted expression in normal tissues, STEAP1 has become an attractive target for next-generation oncology drug development.
STEAP1 is gaining strong attention in advanced prostate cancer, especially metastatic castration-resistant prostate cancer (mCRPC), where there remains a significant need for differentiated targets beyond existing prostate cancer therapies. Its membrane localization makes it highly relevant for antibody-based, cell therapy, and targeted delivery approaches.
🧬 STEAP1 is highly relevant in:
• Prostate cancer
• Metastatic castration-resistant prostate cancer (mCRPC)
• Ewing sarcoma
• Bladder cancer
• Colorectal cancer
• Breast cancer
• NSCLC and other solid tumor research
🎯 Targeting STEAP1 in oncology drug development:
🧪 Antibody-drug conjugates (ADCs) – STEAP1 provides a compelling surface antigen for tumor-selective payload delivery in solid tumors.
🧪 CAR-T cell therapy – STEAP1 CAR-T programs are being explored to address advanced prostate cancer, with preclinical studies showing antitumor activity in metastatic prostate cancer models.
🧪 T-cell engagers / bispecific antibodies – STEAP1-targeted T-cell redirecting strategies are emerging as a promising approach to drive immune-mediated tumor killing.
🧪 Radioligand and imaging strategies – STEAP1 may support new diagnostic, patient selection, and targeted radiotherapeutic approaches in prostate cancer.
🧪 Combination strategies – STEAP1-targeted modalities are being evaluated alongside androgen receptor pathway inhibition and other prostate cancer treatment backbones.
💥 Challenges include:
• Tumor antigen heterogeneity
• Antigen escape and resistance mechanisms
• Solid tumor microenvironment barriers
• Balancing potency with on-target/off-tumor safety
• Translating strong preclinical activity into durable clinical benefit
🔬 Ongoing research is positioning STEAP1 as one of the most promising membrane protein targets in prostate cancer and solid tumor therapeutics. As the field moves beyond single-target prostate cancer strategies, STEAP1 may play an important role in expanding the next generation of ADCs, bispecific antibodies, CAR-T therapies, and precision oncology platforms.
