mRNA-tLNP
In vivo CAR-T is reshaping the future of cancer therapy by enabling T cells to be engineered directly inside the patient, eliminating the need for complex ex vivo cell manufacturing. To achieve this breakthrough, precision delivery is essential. Antibody-conjugated lipid nanoparticles (mRNA-tLNPs) provide a powerful solution, combining the efficiency of mRNA-LNP delivery with the targeting ability of antibodies. By directing genetic payloads specifically to T cells, this technology drives selective in vivo CAR expression, reduces off-target effects, and enhances therapeutic potential. mRNA-tLNPs thus play a pivotal role in making in vivo CAR-T not only possible, but scalable and clinically accessible.

Validation of CD8-Targeted LNP Binding Specificity
After anti-CD8 antibodies were conjugated to LNPs, the resulting tLNPs generated strong and stable targeted fluorescent signals in the CD8 target system, whereas the control group showed markedly reduced signal intensity. In PBMCs, the tLNPs specifically bound to CD8⁺ T cells, with imaging results consistent with flow cytometry data. These findings validate the targeting specificity and system stability of the antibody-conjugated LNP platform.

Antibody conjugation copy number
The LNP antibody conjugation copy number refers to the average number of antibody molecules successfully conjugated to each lipid nanoparticle (LNP).
FAQ
What is targeted LNP (tLNP) and how does it work for mRNA delivery?
Targeted lipid nanoparticles (tLNPs) are advanced delivery systems designed to transport mRNA to specific cell types. Unlike conventional LNPs, tLNPs are functionalized with targeting ligands such as antibodies or peptides that recognize cell surface markers. This enables selective delivery of mRNA to target cells, improving delivery efficiency while reducing off-target distribution.
How can targeted LNP deliver mRNA to T cells or immune cells?
tLNPs can be engineered with ligands that bind to specific receptors on T cells or other immune cells. Once bound, the nanoparticles are internalized through receptor-mediated endocytosis, allowing the encapsulated mRNA to be released into the cytoplasm. This targeted approach enhances delivery precision and is particularly useful for applications such as in vivo CAR-T generation and immune cell engineering.
What are the advantages of targeted LNP over conventional LNP?
Compared to conventional LNPs, targeted LNPs offer improved cell specificity, reduced off-target effects, and enhanced therapeutic efficiency. While standard LNPs tend to accumulate in organs such as the liver, tLNPs can be directed to specific cell populations, enabling lower dosing requirements and more controlled biological responses. This makes them highly valuable for next-generation mRNA therapeutics.
How far can Levostar support the t-LNP projects?
Levostar can provide end-to-end solutions, including payloads synthesis, LNP foumulation choices, antibody conjugation, and final QC.
What is nano flow cytometry?
Nano flow cytometry is an unique QC method desinged by Levsotar. It collects fluorescence intensity to quantify the LNP antibody conjugation positive rate and antibody copy number per LNP. This rapid and precise quantification is essential for t-LNP proejcts.
