Drug Target Encyclopedia Series (24) —CCR8

CCR8 is a G protein–coupled chemokine receptor primarily expressed on tumor-infiltrating regulatory T cells (Tregs).
In cancer immunotherapy, CCR8 has emerged as a highly selective immune checkpoint target, enabling Treg depletion within the tumor microenvironment (TME) while sparing peripheral immune tolerance.

Because of its restricted expression pattern, CCR8 is widely regarded as a next-generation immuno-oncology target.

CCR8 is highly expressed in:

  • Tumor-infiltrating Tregs (TI-Tregs)
  • Solid tumors with immunosuppressive TMEs, including:
    • Non–small cell lung cancer (NSCLC)
    • Breast cancer
    • Colorectal cancer
    • Ovarian cancer
    • Head and neck squamous cell carcinoma (HNSCC)
  • Select inflammatory and allergic disease contexts

High CCR8 expression correlates with immune evasion, poor prognosis, and resistance to checkpoint inhibitors.

Targeting CCR8 in cancer immunotherapy

🧪 Monoclonal antibodies (mAbs)

Anti-CCR8 antibodies are designed to selectively deplete intratumoral Tregs via ADCC, enhancing effector T-cell activity without systemic autoimmunity.

🧪 Combination immunotherapy

CCR8 blockade is being actively explored in combination with:
• PD-1 / PD-L1 inhibitors
• CTLA-4 antibodies
• Cancer vaccines

to convert “cold” tumors into immune-responsive ones.

🧪 Antibody engineering & Fc optimization

Next-generation CCR8 mAbs focus on Fc-engineering to maximize tumor-restricted Treg depletion.

🧪 mRNA & RNA-based technologies

mRNA tools are increasingly used to:
• Study CCR8-mediated Treg biology
• Validate antibody and CAR constructs
• Model CCR8 expression in immune cells
• Support transient expression for safer immunotherapy research

Key challenges

  • Achieving tumor-restricted Treg depletion
  • Avoiding systemic immune activation
  • Limited availability of robust preclinical CCR8 models
  • Translating TME-specific biology into durable clinical responses

Current research directions

  • CCR8 antibodies with enhanced ADCC
  • Combination regimens with PD-1/PD-L1 blockade
  • Biomarker-driven patient stratification
  • mRNA-based immune modeling for rapid target validation
  • Understanding CCR8’s role in TME-specific immune suppression
