BAFF, also known as B-cell Activating Factor or TNFSF13B, is a member of the TNF ligand superfamily and an important immune regulatory molecule involved in B-cell survival, maturation, and antibody production.
BAFF exists as both a membrane-bound protein and a soluble cytokine, interacting with receptors including BAFF-R, TACI, and BCMA to regulate B-cell homeostasis and humoral immune responses. Dysregulated BAFF signaling is closely associated with autoimmune diseases, B-cell malignancies, and chronic inflammatory disorders.
🧬 BAFF is highly relevant in:
• Systemic lupus erythematosus
• Sjögren’s syndrome
• Rheumatoid arthritis
• Multiple sclerosis
• B-cell malignancies
• Autoimmune and inflammatory disease research
🎯 Targeting BAFF in drug development:
- Monoclonal antibodies – Neutralize BAFF activity to reduce abnormal B-cell survival and autoantibody production.
- Receptor-Fc fusion proteins – Block BAFF/APRIL-related signaling pathways by trapping soluble ligands.
- Bispecific or dual-target strategies – Explore BAFF-related pathways together with APRIL, CD20, CD19, or other B-cell targets.
- Cell-based functional assays – Evaluate BAFF-mediated B-cell activation, proliferation, survival, and antibody secretion.
- Autoimmune disease strategies – Modulate BAFF-driven B-cell responses to restore immune balance.
💥 Challenges include:
• Complex biology of membrane-bound and soluble BAFF
• Functional overlap with APRIL signaling
• Disease heterogeneity and biomarker selection
• Balancing B-cell suppression with infection risk
• Optimizing therapeutic format, dosing, and patient stratification
🔬 BAFF is gaining continuous attention as a key B-cell regulatory target in autoimmune and inflammatory disease drug discovery. As antibody, fusion protein, and next-generation immune-modulating strategies continue to evolve, BAFF remains an important target for understanding and controlling pathological B-cell responses.
