Levostar Biotech, a technology-driven enterprise specializing in the development and high-quality production of RNA-based therapeutics, has launched its latest independently developed third-generation mRNA-specific plasmid platform—monoPlasmid. In this exclusive interview, RNAScript invited Dr. Wuxin, founder and CEO of Levostar Biotech, to discuss the development background and potential of this innovative platform.
Q: Why did Levostar Biotech choose to explore cutting-edge technology in the mRNA field?
A: As mRNA technology gains global attention, its application potential has expanded rapidly beyond vaccines to emerging fields such as cell therapy, gene editing, and protein replacement. The surge in investment has further accelerated drug development in this space. However, with rapid industrial growth, we have observed that the foundational research and development of mRNA technology remain relatively weak, with many fundamental drug development challenges yet to be addressed. Our team has accumulated over a decade of experience in synthetic biology and nucleic acid therapeutics, leading us to focus on solving key technical challenges—such as mRNA plasmid heterogeneity—through dedicated innovation.
Q: What is mRNA plasmid heterogeneity, and how does it impact drug production?
A: Unlike other types of plasmids, mRNA plasmids contain a long PolyA sequence, which is crucial for the in vivo stability and efficacy of mRNA therapeutics. However, this long PolyA sequence introduces significant heterogeneity. Our research has shown that during plasmid amplification and serial passaging, the PolyA sequence in mRNA plasmids is highly susceptible to deletions, resulting in plasmid mixtures with varying PolyA lengths. Additionally, the presence of PolyA sequences makes mRNA plasmids more prone to dimerization and multimerization compared to other plasmids. The instability of the PolyA sequence, combined with the formation of plasmid multimers, significantly impacts the uniformity of mRNA plasmids, posing a major technical challenge in mRNA drug development.
Plasmid heterogeneity not only affects drug safety and efficacy but also necessitates additional time and cost for process optimization, substantially increasing production expenses. Recognizing these challenges, we have devoted significant effort to developing the monoPlasmid platform, the first domestically introduced platform specifically designed to address mRNA plasmid heterogeneity.
Q: What are the key technological advantages of the monoPlasmid platform in overcoming mRNA plasmid heterogeneity?
A: The monoPlasmid platform leverages Levostar Biotech’s latest synthetic biology patents, combining the mRNA-specific pLevoD series plasmids with our proprietary engineered bacterial strains. Through a combination of directed evolution and AI-driven selection, we have innovatively modified plasmid control elements. Additionally, by precisely editing specific bacterial strains, we have effectively suppressed the formation of multimers. This approach not only resolves the multimer issue but also ensures long-term stability of the PolyA tail across multiple generations of plasmid passaging.
Traditional approaches to addressing plasmid multimerization require extensive screening and fermentation condition optimization for each specific mRNA plasmid and bacterial strain. This process often needs to be repeated for every production batch and lacks a guaranteed success rate. In contrast, plasmids produced by our monoPlasmid platform exhibit exceptionally high homogeneity, eliminating the need for screening and process optimization. This reduces process development time by 50% while ensuring PolyA tail stability. Furthermore, the monomeric supercoiled plasmid ratio exceeds 95%, completely resolving the multimerization problem. Moreover, the reduction in multimers significantly enhances plasmid yield. Without any additional process optimization, the monoPlasmid platform already achieves an mRNA plasmid yield of up to 500 mg/L, which is 2-3 times higher than the average yield of conventional mRNA plasmids.
Q: From your perspective, how will the monoPlasmid platform transform mRNA drug development?
A: The ability to rapidly, stably, and cost-effectively produce mRNA therapeutics is crucial for various applications, such as personalized mRNA vaccines for cancer and cell therapy, where each patient requires a unique mRNA treatment under time-sensitive conditions. Currently, the heterogeneity of mRNA plasmids significantly extends patient wait times and, in some cases, prevents patients from receiving treatment in time. The introduction and application of the monoPlasmid platform will greatly shorten mRNA drug production timelines.
Looking ahead, we hope that more companies, both domestically and internationally, will adopt the monoPlasmid platform to lower production costs, enhance drug quality, shorten manufacturing cycles, and establish more comprehensive quality control systems. By doing so, we can collectively drive technological innovation across the industry.
Q: Beyond continuous research and innovation in the mRNA field, Levostar Biotech is also exploring emerging RNA technologies. Can you share more insights?
A: Absolutely. One of Levostar Biotech’s core competencies lies in our deep expertise in synthetic biology. Building on this foundation, we are continually expanding our research scope. We have already extended our capabilities beyond traditional plasmid and mRNA technologies to develop innovative RNA therapeutics such as circRNA and saRNA. In the future, we plan to gradually introduce these new platforms to the market, aiming to support more pharmaceutical companies with cutting-edge solutions tailored to their needs.
